ABSTRACT

RNA has received m ore attention in the field of forensic m edicine and the developm ent of the new biological m arkers based on RNA show s great significance in the analysis of com plex cases. circular RNA (circRNA ) is a kind of non-coding RNA w hich is w idely reported recently. A lthough the regulatory m echanism s of generation and expression are not fully clear, the existing research indicates that circRNA has im portant biological functions. C ircRNA has a cell-type-specific expression w ith great stability and a high expression level, w hich m akes it m eaningful in forensic applications potentially. In this paper, the research progress, the generation and regulation of circRNA as w ell as its biological characteristics and functions are sum m arized, w hich w ill provide references for related studies and foren-sic applications.

T his article review s several candidate genes and advances in epigenetics w hich are associated w ith violent behavior. T he prospects and significance of violent behavior research from the view of gene polym orphism and epigenetics are also discussed.

ABSTRACT

Objective To analyze the cases of anaphylactic death cases and explore the standards of judi-cial expertise of anaphylactic death for providing evidence for judicial expertise. Methods Fifty-nine cas-es death due to allergic reaction in Shanghai were collected. And details of medical history, clinical manifestation of anaphylactic reaction and postm ortem exam ination findings were review ed for all cases. Results In the 59 cases, there were 58 cases died from drug allergy, including 77.6% of them were an-tibiotics. The rates of treating in standard hospital and illegal clinic were 37.3% and 61.0%, respectively. The allergic sym ptom s were dyspnea and facial cyanosis. The time from contacting allergens to death ranged from 1 m in to 3 d. The concentration of total serum IgE ranged from 50 to 576.92 IU/m L . The results of clinical manifestation and pathological anatomy had obviously changes. Conclusion B ased on the exclusion of all other cause of death and synthetically analysis of details of cases, medical history, clinical manifestation and anatomy, the conclusion of anaphylactic death can reached. The details of cas-es including clinical history, exposure to allergens, and clinical manifestation play an im portant role in diagnosis of anaphylactic death.

ABSTRACT

Objective To explore the relationship between the expression of EIIIA + fibronectin in incised wound of rat’s skin and injury time. Methods The wounding model was established by cutting the dor-salskin of 48 adult SD rats. The rats were sacrificed atthe pre-setinjury time as immediately, 0. 5h, 1h, 2 h, 3 h, 4 h, 6 h, and 8 h. The skin sam ples were taken at the m argin of wound. The expression of the EIIIA + fibronectin was detected by im m unohistochem istry and W estern blotting and the relationship be-tween its expression and injury time was observed. Results The expression of EIIIA + fibronectin was not observed im m ediately. The basal cell of skin began to showpositive expression 0. 5 h after injury. W ith the extension of injury time, positive staining became stronger. The value of relative optical density was gradually increased with prolonged injury time by the W estern blotting analysis. Conclusion The expres-sion of EIIIA + fibronectin could be used for estimation of injury time in the early stage of skin injury.

ABSTRACT

Objective To explore the relationship between renin-angiotensin system (RAS) blockers and plasma lev-el of adiponectin (APN) in patients with metabolic syndrome (Mets). Methods Fifty-three patients with Mets were random-ized into control group (n=23), angiotensin-convertingenzyme inhibitor (ACEI) group (benidipine, 5 mg/d, n=15) and angio-tensinll recepter blockers(ARB)group (candesartan, 4 mg/d, n=15),and were given 4-week treatment. Twenty-three pa-tients, who were avoiding application related drugs affecting the RAS for four weeks, were used as control. The values of plas-ma APN, fasting blood glucose (FBG), fasting insulin (FINS), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), creatinine(CR), aspartate aminotransferase (ALT), bil-irubin (BIL) and insulin resistance index (HOMA-IR) were detected and compared before and after treatment. Results The levels of FBG and LDL-C were significantly decreased after treatment than those before treatment in ACEI group. The level of CR was significantly lower after treatment than that before treatment in ARB group (P<0.05). The plasma APN level was significantly higher after treatment compared to that before treatment in ACEI and ARB groups (P<0.01).There were no sig-nificant differences in the values of FINS,HOMA-IR,TC,ALT and BIL between 'before and after treatment' in three groups (P>0.05). The levels of CR, TC, ALT and BIL before treatment were influencing factors for the APN level. The levels of FBG and TC after treatment were influencing factors for the APN level. Conclusion APN may serve as potential therapeu-tic targets for treating Mets and its related complications by ACEI and ARB.

ABSTRACT

Objective: To study the effects of triptolide-medicated serum on secretory function of adrenocortical cells isolated from rats. Methods: Thirty SD rats were randomly divided into control group, prednisone group, and low-, medium- and high-dose triptolide groups. Rats were administered with normal saline, prednisone and low-, medium- and high-dose triptolide respectively by gastrogavage to prepare sera containing drugs. Primary adrenocortical cells were isolated from normal male rats and cultured with sera containing drug for 48 hours. Expression of proliferating cell nuclear antigen (PCNA) was observed by immunohistochemical method and number of PCNA-positive cells was counted. Ultrastructure of adrenocortical cells was observed under a transmission electron microscope. Content of corticosterone in supernatant of adrenocortical cell culture was detected by enzyme-linked immunosorbent assay, and real-time fluorescence quantitative polymerase chain reaction (PCR) was employed to investigate the expression of 3beta-hydroxysteroid dehydrogenase (3beta-HSD) mRNA. Results: As compared with the control group, content of corticosterone in supernatant of adrenocortical cell culture and expression of 3beta-HSD mRNA were significantly increased in the triptolide-treated groups, and the numbers of PCNA-positive cells were increased in the medium- and high-dose triptolide groups, however, they were decreased in the prednisone group. Conclusion: Triptolide-medicated serum can increase the secretion of corticosterone in rat adrenocortical cells in vitro.

ABSTRACT

Objective To investigate the expression of sLc22A1 8,an impfinted tumor suppressor gene,in breast cancer and explore the relationship between expression of SLC22A18 and the pathogenesis of breast cancer. Methods Real-time quantitative reverse transcriptase-polymerase chain reaction was applied on 46 cases of infiltrating duetal breast carcinoma(IDC),46 csges of corresponding adjacent noncancerous tissues and 20 benign breast tissues in order to detect mRNA expression of SLC22A18 gene.Protein expression was detected by immunohistochemistry.Statistical analysis was carried out to analyse the correlation between SLC22A18 gene expression and various elinical parameters in these breast cancer patients. Results SLC22A18 mRNA expression in 46 IDC tissues Was lower than that in all corresponding adjacent non-cancerous tissues(Z=-4.900,P<0.01).SLC22A18 mRNA expression was lower in breast cancer eases,when compared with that in benign cases(Z=-3.182,P<0.01).SLIC22A18 mRNA expression in 40 IDCs Was lower than that in 6 dutal carcinoma in situ(part of IDC)(Z=-2.022,P<0.05).There was a decreased or completely diminished SLC22A18 protein expression in breast cancer.A significant difference of SLC22A18 protein expression was also observed in IDC and benign groups(P<0.05).Neither mRNA nor protein expression of SLC22A18 gene correlated with clinieopathologic parameters such as age of patients,size of tumor,ehnical stage,pathologic subtype,histologlc grade or lymph node metastasis(P>0.05).Condusion Decreased expression of SLC22A18 gene may play an important role in the carcinogenesis of IDC.

AIM:To study the mRNA expression and methylation status of imprinted gene SLC22A18 in infiltrating ductal carcinomas(IDCs),and the correlation between methylation status and clinical characteristics in IDCs.METHODS:The methylation status at the promoter regions of SLC22A18 gene was examined by methylation-specific polymerase chain reaction(MSP)in the specimens of IDC from 40 patients.The mRNA expression of SLC22A18 gene was detected by real-time reverse quantitative transcriptase polymerase chain reaction(real-time RT-PCR)in 40 specimens of IDC and the cell line MDA-MB-231.The cell line MDA-MB-231 was treated with 5-aza-2'-deoxycytidine(5'-aza-dc)and trichostatin A(TSA),then MSP and rea1-time RT-PCR were used to detect the methylation status and mRNA expression levels of SLC22A18 gene.RESULTS:SLC22A18 mRNA expression in 40 IDC tissues was lower than that in al1 corresponding adjacent non-cancerous tissues(P

The fibronectin was purified from human plasma by Gelatin-Sepharose 4B affinity chromatography. The rabbits were immunized with the fibronectin. the raw anti-fibronectin sera were absorbed bythe solid phase of the fibronectin free human serum. Then the specific anti-fibronectin sera were obtained. ABSTRACT

Six cases of suicidal sedative-soporific drug poisoning is reported.The main clinical features were coma,respiratory depression,hypotension,disappearance of reflexes and peripherel circulatory failure.Thepathological fingdings were as follows:pulmonary congestion and edema;fatty degeneration of the liver;degeneration of the epithalial cells of the proximal convoluted tubules and cerebral edema.The severityof the pathological changes depend upon the duration between the drug administration and the time of deathThe longer the course,the more prominent the changes.The fatality rate related with the dose of drugadministration,the underlying deseases and the complications.
